Blue Stained
After performing the acid-fast staining procedure on M.smegmatis, the cells stained blue, what went wrong?
I totally don't get this one. Could it be that I left the stain on for too long?
This is probably the case. I don't know if they teach you the paper towel method in school or what, but I have a much simpler one for you for next time. Put the carbol fuschion onto the slide and flame with your bunsen burner (not allowing the liquid to boil) for 1.5 minutes, then allow the stain to sit for another 1.5 minutes. Rinse thoroughly, yet gently with water. Apply the acid alcohol and let sit for 2 minutes, rinse again, apply the counterstain for about 2 minutes and rinse. Your slides should turn out much better!
